PNC-27 is a synthetic membrane active peptide derived from the HDM-2 binding domain of p53 (amino acids 12-26), linked to a transmembrane penetrating leader sequence from the Drosophila antennapedia homeodomain protein. Originally developed using a supercomputer at SUNY Downstate Medical Center in 2000, PNC-27 interacts with the MDM2 p53 tumor suppressor complex, acting as a competitive inhibitor to increase the half-life of p53 and facilitate the elimination of cancer cells. Hudson Science’s formulation supplies 10mg as a lyophilized vial, preserving peptide structural integrity across storage conditions until reconstitution.
PNC-27’s mechanism of action involves binding to the HDM-2 protein, which is overexpressed in many cancer cell membranes by binding to membrane bound HDM-2, PNC-27 creates oligomeric pores in the cancer cell plasma membrane, leading to cancer cell necrosis through membrane lysis. A notable aspect of PNC-27’s research profile is its ability to induce tumor cell necrosis independently of the p53 pathway. A significant characteristic because many cancer cells have compromised or mutated p53 which can render other treatments less effective, making PNC-27’s p53 independent action a research differentiator applicable to a wider range of tumor types.
Untransformed or non cancerous cells do not appear to form oligomeric pores when exposed to PNC-27, with research suggesting this selectivity is caused by MDM2’s mis localization to cancerous cell plasma membranes. This selective activity profile has generated significant research interest in PNC-27 as a tool compound for studying cancer cell membrane biology, HDM-2 expression patterns across tumor types, and the p53 independent mechanisms of tumor cell elimination. PNC-27’s evidence base remains predominantly preclinical; it is not an approved cancer treatment and human clinical data is limited at this stage of research.
