Rybelsus contains semaglutide, a glucagon like peptide 1 (GLP 1) receptor agonist used in adults with type 2 diabetes mellitus alongside diet and exercise to improve glycemic control. The 9mg tablet belongs to the newer optimized Rybelsus formulation introduced in Canada and is currently listed by Health Canada as a marketed prescription product.
Semaglutide is structurally similar to the naturally occurring hormone GLP-1, which is released by the gastrointestinal system following food intake. By activating GLP-1 receptors, semaglutide increases glucose dependent insulin secretion, reduces inappropriate glucagon secretion and slows gastric emptying. Together, these effects help improve blood glucose regulation while also influencing appetite and food intake.

In Canada, Rybelsus is indicated as an adjunct to diet and exercise to improve glycemic control in adults with type 2 diabetes. Semaglutide also carries important precautions relating to pancreatitis, gallbladder disease, dehydration and kidney complications, diabetic retinopathy and hypoglycemia when combined with insulin or insulin secretagogue medications.
Canadian Rybelsus prescribing information also contains a serious warning concerning thyroid C cell tumours observed in animal studies. It is contraindicated in individuals with a personal or family history of medullary thyroid carcinoma (MTC) or Multiple Endocrine Neoplasia syndrome type 2 (MEN 2). Whether semaglutide causes thyroid C cell tumours in humans remains uncertain.
